Quantification of Fusarium commune in Douglas-fir Seedling Nurseries

A better diagnostic assay can help growers make cost effective management decisions that meet environmental regulations. A quantitative real-time polymerase chain reaction (qPCR) assay has been developed for both Fusarium commune and Fusarium oxysporum isolated from inoculated soil, with the intent of quantifying F. commune from naturally occurring populations in nursery soil. F. commune inoculated in greenhouse potting soil (CFU/g) positively correlated with the observed Ct values from the duplex qPCR assay. Additionally, greenhouse trials show a positive correlation between inoculum addition and disease development at high levels of inoculation. Additional trials are being done.
